Does it do it at the same speed in 4th ?? if it does it's not likely to be the engine... or does it do it at the same revs in forth ?? in which case it might be the engine, if it doesn't do it at all in forth then it's more than likely the gearbox or other drivetrain issue...

I once had a similar sounding problem on a Fiat Coupe - slight misfire at a specific rpm, only happened on very light throttle, turned out to be a coil pack problem
